Article VI of the U.S. Constitution

For all you law experts (or soon to be), article VI declares the Constitution to be the Supreme Law of the Land (Supremacy clause). What effect, if any, does this clause have, where a state law directly conflicts with a federal law? Which law will prevail, and why?
